AGO2
AGO2,全稱為Argonaute 2,也常被稱為EIF2C2,是RNA誘導(dǎo)沉默復(fù)合體(RISC)的核心組成部分。它在細(xì)胞內(nèi)的基因調(diào)控網(wǎng)絡(luò)中占據(jù)關(guān)鍵位置,能與微小RNA、小干擾RNA等小分子核酸緊密結(jié)合,精準(zhǔn)識別特定靶基因序列,進(jìn)而調(diào)控這些基因的表達(dá)水平,參與細(xì)胞增殖、分化、凋亡及代謝等多種基礎(chǔ)生命活動的調(diào)節(jié)。
在疾病領(lǐng)域,AGO2的異常表達(dá)或功能失調(diào)與多種病癥相關(guān):部分惡性腫瘤(如肺癌、乳腺癌)中,它的表達(dá)量明顯上升,可能推動腫瘤細(xì)胞的侵襲與轉(zhuǎn)移;此外,它在機體抗病毒防御中發(fā)揮作用,一些病毒會通過抑制AGO2的活性來逃避宿主免疫監(jiān)控。目前,針對AGO2的藥物研發(fā)尚處于早期探索階段,研究團(tuán)隊正致力于開發(fā)能調(diào)節(jié)其功能的小分子化合物或核酸類制劑,期望通過靶向這一蛋白來修復(fù)紊亂的基因調(diào)控,為癌癥、慢性病毒感染等疾病開辟新的治療方向。

Immunofluorescence staining of MCF-7 with CSB-RA965823A0HU at 1:30, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 492-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
Overlay Peak curve showing Hela cells stained with CSB-RA965823A0HU (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1μg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1μg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
