Wnt3a
Wnt3a即Wnt家族成員3A，是一類分泌型糖蛋白，在機體發(fā)育和細胞生理過程中發(fā)揮關(guān)鍵作用。它通過與細胞膜上的卷曲蛋白受體及低密度脂蛋白受體相關(guān)蛋白5/6結(jié)合，穩(wěn)定細胞質(zhì)中的β-連環(huán)蛋白，使其進入細胞核調(diào)控靶基因表達，同時也能參與調(diào)控非經(jīng)典相關(guān)通路，對細胞增殖、分化及胚胎正常發(fā)育起到重要調(diào)控作用。該蛋白異常會引發(fā)多種疾病，過度表達時會促進結(jié)直腸癌、乳腺癌、胰腺癌等腫瘤的發(fā)生發(fā)展，而表達不足則可能導(dǎo)致胚胎發(fā)育畸形。目前相關(guān)治療研發(fā)圍繞其展開，IWP-2等小分子物質(zhì)可抑制其分泌，處于前期研究階段；抗Wnt3a中和抗體、低密度脂蛋白受體相關(guān)蛋白6拮抗劑在動物實驗中展現(xiàn)出抗腫瘤效果，多款靶向Wnt3a的治療方案正針對實體腫瘤進行臨床評估，有望為相關(guān)疾病治療提供新方向。

IHC image of CSB-PA15549A0Rb diluted at 1:260 and staining in paraffin-embedded human breast cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
