SIRT3
SIRT3,作為線粒體中的關鍵去乙酰化修飾酶,其結構包括一個催化區以及調節酶活性的N端和C端結構。它在細胞應激時轉移到線粒體內并成熟,從而發揮其去乙酰化功能。SIRT3主要通過調節線粒體結構和功能,如激活長鏈脂酰輔酶A脫氫酶,在脂肪代謝中發揮重要作用。在信號通路方面,SIRT3通過調控GSK-3β/β-catenin信號通路來抑制NLRP3炎性小體的表達,從而保護骨骼成骨。同時,在腎臟疾病中,SIRT3通過去乙酰化作用抑制腎纖維化,并調控能量代謝穩態。此外,SIRT3還參與心臟氧化應激、缺血再灌注損傷等生理和病理過程。
