OFD1
OFD1 ,全稱為 Oral-facial-digital syndrome 1 protein,常見別名包括 71-7A、CXorf5、JBTS10、RP23、SGBS2 等。OFD1 蛋白在細胞中主要位于中心體和中心粒衛星區域,參與初級纖毛的生物合成過程。初級纖毛是細胞表面的微管結構,在多種信號轉導過程中發揮重要作用,對于細胞的發育和組織的穩態維持至關重要。OFD1 蛋白通過調節 Wnt 信號通路和左右軸的指定,在發育過程中發揮重要作用。此外,OFD1 蛋白還參與纖毛生成過程的調控,其缺失或錯義突變會導致初級纖毛的喪失,從而引發多種疾病。
由于 OFD1 蛋白與多種疾病相關,因此其成為藥物研發的重要靶點。目前,針對 OFD1 蛋白的藥物研發主要集中在口服-面部-指綜合癥和 2 型 Simpson-Golabi-Behmel 綜合癥的治療上。研究人員通過基因編輯技術構建了 OFD1 蛋白突變的小鼠模型,用于研究該蛋白突變對細胞功能的影響,并探索潛在的藥物靶點。此外,還有研究關注 OFD1 蛋白與其他蛋白的相互作用,以及其在不同疾病中的作用機制,以期開發更精準的藥物治療方法。目前,已有一些針對 OFD1 蛋白的藥物進入臨床研究階段,期待未來能有更多有效藥物問世,為患者帶來新的治療選擇。

IHC image of CSB-PA016289LA01HU diluted at 1:100 and staining in paraffin-embedded human skeletal muscle tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at 37℃ Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-Mouse IgG labeled by HRP and visualized using 0.05% DAB.
