ITAM
ITAM蛋白，全稱為免疫受體酪氨酸激活基序（Immunoreceptor tyrosine-based activation motif），是一種磷酸化基序，常見別名包括免疫受體酪氨酸激活基序。ITAM蛋白主要參與免疫細(xì)胞的信號傳導(dǎo)，其酪氨酸殘基被磷酸化后，能夠招募胞內(nèi)蛋白激酶或銜接蛋白，從而傳遞活化信號，促進(jìn)細(xì)胞活化。ITAM蛋白廣泛存在于免疫受體中，如T細(xì)胞受體復(fù)合物、B細(xì)胞受體復(fù)合物以及某些Fc受體，參與免疫細(xì)胞的發(fā)育、分化和功能調(diào)節(jié)。
ITAM蛋白與多種疾病相關(guān)，如免疫缺陷病、自身免疫病、腫瘤等。例如，ITAM蛋白的突變或功能異常可能導(dǎo)致T細(xì)胞或B細(xì)胞的功能缺陷，進(jìn)而引發(fā)免疫缺陷病。此外，ITAM蛋白還與腫瘤的發(fā)生發(fā)展密切相關(guān)，其異常表達(dá)或功能可能導(dǎo)致腫瘤細(xì)胞的增殖、轉(zhuǎn)移和耐藥性。
目前，針對ITAM蛋白的藥物研發(fā)主要集中在免疫調(diào)節(jié)方面。例如，研究人員正在開發(fā)能夠抑制ITAM蛋白磷酸化的藥物，以抑制免疫細(xì)胞的活化，從而治療自身免疫病和腫瘤等疾病。此外，研究人員還在開發(fā)能夠特異性地靶向ITAM蛋白的抗體藥物，以治療免疫缺陷病和腫瘤等疾病。
