GPR20
GPR20，全稱為G protein-coupled receptor 20，是一種在多種組織中表達(dá)的孤兒GPCR。該蛋白在胃腸道間質(zhì)瘤（GIST）中高度表達(dá)，成為治療GIST的非激酶新靶點(diǎn)。研究表明，GPR20在GIST細(xì)胞系和患者來(lái)源異種移植（PDX）模型中表達(dá)顯著增加。基于這一發(fā)現(xiàn)，研究者開發(fā)了靶向GPR20的抗體藥物偶聯(lián)物（ADC）DS-6157a，該藥物在臨床前模型中表現(xiàn)出依賴GPR20表達(dá)的抗腫瘤活性，對(duì)TKI耐藥的GIST有效。DS-6157a的臨床前評(píng)估顯示其對(duì)TKI耐藥的GIST模型具有顯著抗腫瘤活性。此外，早期臨床試驗(yàn)表明，DS-6157a的藥代動(dòng)力學(xué)特性與安全性之間存在明顯的關(guān)系，藥物暴露量與中性粒細(xì)胞減少癥和血小板減少癥等血液學(xué)不良事件相關(guān)。盡管DS-6157a的研發(fā)已被取消，但第一三共擁有強(qiáng)大的ADC藥物管線，包括Enhertu等。
