DKK1

DKK1，全稱為Dickkopf-related protein 1，別名包括SK、DKK-1等。該蛋白是一種分泌型糖蛋白，主要由成骨細胞和骨細胞分泌，也表達于胎盤、皮膚、前列腺等多種組織和細胞類型。DKK1通過與Wnt蛋白競爭性結合LRP5/6受體，拮抗Wnt/β-catenin信號通路活性，從而調節細胞增殖、分化及癌變，影響細胞凋亡、瘤細胞侵襲和轉移。研究表明，DKK1在不同細胞中表現出相反的調節作用，其功能相對復雜。
在腫瘤發生發展中，DKK1的作用呈現多樣性。在某些腫瘤中，如多發性骨髓瘤、肝細胞癌、非小細胞肺癌等，DKK1表達增高，呈現促癌基因的特征，促進腫瘤生長和轉移。而在另一些腫瘤中，如胃癌、膀胱癌、黑色素瘤等，DKK1表達降低，呈現抑癌基因特征，抑制腫瘤生長和轉移。此外，DKK1還與帕金森病、強直性脊柱炎、牙周炎等多種疾病有關。
目前，針對DKK1的藥物研發取得了一定的進展。多種DKK1抑制劑和調節劑處于臨床前或藥物發現階段，用于治療肝癌、卵巢癌、子宮內膜癌、前列腺癌等多種疾病。
