The first step of making this recombinant Mouse Nid1?protein is synthesizing the Nid1?gene, the synthesized gene is then cloned into an expression vector which is a cDNA plasmid that includes a promoter sequence and an antibiotic-resistant gene. It also has Tag-Free?encoding a fusion tag for downstream protein purification or identification. The antibiotic-resistant gene enables the selection of cells carrying the plasmid in antibiotic-based media and then transfects cells with a DNA vector that contains the template and then culturing the cells so that they transcribe and translate the desired protein. The recombinant Nid1 protein is purified by ion-exchange chromatography or affinity purification. And the purity is 90%+ by SDS-PAGE.
Nid1, also known as entactin-1, is a glycoprotein found in basement membranes. It functions as a linker between laminins, collagens, and proteoglycans in the extracellular matrix and binds to cell surface integrins, assisting in the establishment and maintenance of the basement membrane and tissue architecture. Nid1 also regulates cell polarization, migration, and invasion through interactions with cell receptor molecules. Braconi et al. reported that Nid1 is upregulated in hepatocellular cancer with vascular invasion by whole-genome analyses. Zhang Y et al. discovered that Nid1 was an ovarian cancer-associated protein and can be detectable in peripheral blood, with increased expression in ovarian cancer patients. Later, Nid1 was found to hold the potential to be a helpful biomarker in the diagnosis of ovarian serious carcinoma.
